چکیده

Several studies indicate that the nuclear factor-kappa B (NFB) -activation cascade plays a crucial role not only in immune responses, inflammation, and apoptosis but also in the development and processing of pathological pain. Accordingly, a pharmacological intervention into this pathway may have antinociceptive effects and could provide novel treatment strategies for pain and inflammation. In this review we summarize the role of NFB in the nervous system, its impact on nociception, and several approaches that investigated the effects of various modulators of the classical IBkinase-NFB signal transduction pathway in inflammatory nociception and neuropathic pain. The results indicate that NFB has an impact on nociceptive transmission and processing and that a number of substances that inhibit the NFB-activating cascade are capable of reducing the nociceptive response in different animal models. Therefore, a modulation of specific participants in the NFB signal transduction might exert a useful approach for the development of new painkillers.—Niederberger, E., Geisslinger, G. Neuroinflammatory disorders include a wide spectrum of disorders in which immune system injures components of the nervous system. Despite the advances in therapy, lack of efficient curative therapies for these disorders not only affects the quality of life but also places huge burden on the society. These limitations have necessitated therapeutic interventions for developing more efficient regi...

Modulation of IKKβ/NF-κB and TGF-β1/Smad via Fuzheng Huayu recipe involves in prevention of nutritional steatohepatitis and fibrosis in mice

Objective(s):Fuzheng Huayu recipe (FZHY) exerts significant protective effects against liver fibrosis by strengthening the body’s resistance and removing blood stasis. However, the molecular mechanisms through which FZHY affects liver fibrosis are still unclear. In this study, we examined the expression levels of factors involved in the inhibitor κB kinase-β (IKK-β)/nuclear factor-κB (NF-κB) an...

RNAi technology: A Novel approaches against fungal infections

Despite the introduction of new antifungal agents, resistances to antifungal therapy continue to increase and outcome of invasive fungal infections treatment is frequently suboptimal. A large amount of the recent effort in antifungal drug discovery has focused on a limited set of targets with functions known or expected to be important for fungal viability and virulence. A variety of techniques...

Tumor necrosis factor alpha inhibition of hepatitis B virus replication involves disruption of capsid Integrity through activation of NF-kappaB.

Chronic infection by hepatitis B virus results from an inability to clear the virus, which is associated with liver disease and liver cancer. Tumor necrosis factor alpha (TNF-alpha) is associated with noncytopathic clearance of hepatitis B virus in animal models. Here we demonstrate that the nuclear factor kappaB (NF-kappaB) signaling pathway is a central mediator of inhibition of hepatitis B v...
